Topics to be covered include:

  • Basics and advance techniques in Laser Scanning Confocal Microscopy (LSCM), image formation, 3-D reconstruction, and fluorphores labelling conditions for various biological applications.

  • Selection of filters for confocal fluorescence imaging, various immuno-cytochemical techniques, photo bleaching and data analysis.

  • Localization of proteins in live and fixed cells

                                                                                        

  • Introduction of SPM, basic principles of STM and AFM, AFM equipment description, operating modes, AFM probes, etc.

  • SPM applications in biological and life science

  • Practical session: sample preparation, AFM set-up, imaging in contact and tapping mode, high resolution imaging of selected samples (nanoparticles, carbon nanotubes, biomolecules and soft surfaces)

  • SPM beyond imaging: characterization of chemical composition, interfacial surface properties, nanomechanical properties (nanoindentation), nanolithography etc.

  • A new developments in SPM: NSOM, combined confocal and Raman AFM

  • Practical session: probing of surface properties by force measurements (examples self-assembled monolayers, protein layers, and polymers).

                                                                                        

  • Basics and advances of High-Resolution Transmission Electron Microscope (HR-TEM).

  • Obtaining HR-TEM images and analysing it using crystallographic softwares.
